...... Is this the first meeting of two premiership teams that have played against each other in the other 3 senior tiers of English football, I'm not sure just wondering.

Interesting question. I don't have the answer, but do wonder if Brighton and Swansea, or Bournemouth and Swansea hold the accolade.

Hull and Swansea must have as well? Got no facts to back it up but I believe they've all been in the three EFL leagues together just this last 15 years or so.
Then again I could be wrong as Hull went up into the Prem the first time when Swansea went up to the Championship in 2008, we were at Wembley the day after Windass' famous winner of course. I don't know anyway.

Bournemouth would have played Swansea in the 02-03 season when SO'D got Bournemouth promoted via the play offs and Swansea won to stay up on the final day of the season.

Hull would have also played Swansea more recently in the 03-04 season when we won the league.

I think the answer is probably Coventry versus Crystal Palace (following taken from the great 11v11 site)
1958-59 Division 4
1961-62 Division 3
1964-65 Division 2
1992-93 Premier League (first season of PL)
By the way they also played in the old Division One starting in 1969-70, and even played in the pre 1958 Division 3 South and the old Southern league First Division prior to WW1
By the way I wrote an article for the fanzine in 2012 covering all the (then) teams that had played in all 4 divisions
Edit: Amazingly that is not the answer, or even second!
Coventry City, Crystal Palace and Oldham all played each other in the inaugural Premier League season, having played each other earlier at all 3 lower levels. The fixtures of 1992-93 had Oldham playing Palace on 19 August 1992, Oldham versus Coventry on 5 September 1992 and Coventry versus Palace on 3 October 1992.
So Oldham versus Crystal Palace is the answer.
